摘要:

蛋白O-糖苷键连接的N-乙酰葡萄糖胺(O-GlcNAc)修饰是一种广泛存在的、动态的转录后修饰方式, 通过N-乙酰氨基葡萄糖转移酶(OGT)及N-乙酰氨基葡萄糖苷酶(O-GlcNAcase)将N-乙酰葡萄糖胺单糖添加或移除到蛋白质的丝/苏氨酸残基上,在许多生化过程中起到重要作用。近几年大量研究证明,短时间内O-GlcNAc修饰水平的提高能产生显著的心血管保护作用,在缺氧-复氧及感染性休克刺激时尤为显著,主要可能机制包括抗炎症反应,促热休克蛋白生成,抑制细胞凋亡等。

Abstract:

O-linked attachment of the monosaccharide-N-acetylglucosamine (O-GlcNAc) is a highly dynamic and ubiquitous posttranslational protein modi- fication, plays a critical role in regulating numerous biological processes. O-GlcNAc transferase and O-GlcNAcase are responsible for the addition and removal of O-GlcNAc from serine/threonine residues, respectively. Recently, a number of studies demonstrating that acute increases in O-GlcNAc synthesis afford remarkable cardiovascular protection, especially against hypoxia-reoxygenation and septic stress stimuli, main potential mechanisms include:anti-inflammatory, increased synthesis of HSPs, anti-apoptosis.
