摘要:

疼痛是临床上常见却又难以达到满意治疗效果的综合征。以往的镇痛药物主要致力于阻断疼痛刺激向大脑传递,而非针对疼痛产生的根源,即介导疼痛的受体。目前随着疼痛刺激性受体的识别与克隆,疼痛治疗领域得以新的发展。瞬时感受器电位(transient receptor potential,TRP)就是参与多种生理过程的通道蛋白。瞬时感受器电位香草酸受体1(TRPV1)是TRP中的一员,是一种非选择性阳离子通道,自1997年被克隆以来,很多资料证明了它参与炎性痛和神经病理性痛的产生与发展。TRPV1主要表达于伤害性神经元,它控制着一些神经肽和递质的释放,产生的动作电位被传送到高级神经中枢而产生痛觉。它的激动剂和阻断剂,通过不同方式阻断疼痛的产生与传导,从而达到药物靶向镇痛的目的。

Abstract:

Pain is difficult to cure satisfactorily.The analgesic drugs in the past have focused on preventing the transmission of action potentials from reaching the brain rather than pinpointing the springhead giving birth to pain:the receptor itself.But now it has mostly changed.Transient Receptor Potential (TRP) channels have been implicated in several physiological processes.Transient Receptor Potential vanilloid 1(TRPV1) is one of the family of the TRPs.In recent years,compelling data has been gathered on the role of this channel in inflammatory and neuropathic states.The agonist and antagonist of it interrupt the generation and transmission to achieve the purpose of analgesia.
