摘要:

围手术期神经认知障碍(PND)作为一类常见的麻醉手术后神经系统并发症,严重影响患者术后转归。PND的确切致病机制目前尚不清楚,但最近研究显示,同源性磷酸酶张力蛋白诱导激酶1(PINK1)/帕金森蛋白(Parkin)线粒体自噬通路在不同麻醉药物和方法所致PND的发生、发展过程中具有潜在重要调控作用。文章介绍了PINK1/Parkin线粒体自噬通路,列举了常见的麻醉药物和麻醉方法通过氧化应激和(或)神经炎症等途径参与调控PINK1/Parkin线粒体自噬通路,为PND的防治提供新的治疗靶点和思路。

Abstract:

Perioperative neurocognitive disorders (PND), as a common type of neurological complication after anesthesia and surgery, seriously affect the postoperative outcomes of patients. Although the exact pathogenic mechanism of PND is currently unclear, recent studies have shown that the homologous phosphatase tensin induced kinase 1 (PINK1)/Parkinson's protein (Parkin) mitochondrial autophagy pathway has a potentially important regulatory role in the occurrence and development of PND caused by different anesthetic drugs and methods. Therefore, this review introduces the PINK1/Parkin mitochondrial autophagy pathway and lists common anesthetic drugs and methods that participate in regulating the PINK1/Parkin mitochondrial autophagy pathway through oxidative stress and/or neuroinflammation. It can provide new therapeutic targets and directions for the prevention and treatment of PND.
