Propofol promotes proliferation and up-regulates phosphorylation of cAMP-response element binding protein in adult neural stem cells

摘要:

目的 观察异丙酚对体外培养的大鼠成年神经干细胞是否具有生长调控作用及可能的分子机制。方法 大鼠海马成年神经干细胞接种贴壁后,模拟临床应用浓度加入异丙酚至终浓度10、25、50、100 μg/ml,溶剂(脂肪乳)为对照,培养24 h。用DAPI染核,检测细胞数目变化。用免疫印记法检测细胞CREB磷酸化水平变化。结果 异丙酚(>10 μg/ml)处理组比对照组细胞数显著升高,且呈剂量依赖性。异丙酚处理前后,细胞均表达CREB和磷酸化CREB,但处理组比对照组的CREB磷酸化水平显著升高。结论 异丙酚可能是通过上调细胞中CREB的磷酸化水平从而促进大鼠海马成年神经干细胞的增殖能力。

Abstract:

【Abstract】Objective To study the effects of propofol on the regulation of rat adult neural stem cells (ANSCs) proliferation and to explore the possible molecular mechanisms. Methods Propofol with clinically relevant concentrations (10, 25, 50, 100 μg/ml) was applied into the cultured rat adult neural stem cells media. The vehicle solution of intralipid was as the control. After 24 hours of incubation, cell nuclei were dyed with DAPI to check the variance of proliferation and apoptosis. Westernblot was exploited to detect the phosphorylation level of CREB. Results Propofol promoted the proliferation of ANSCs in a dose-dependent manner. CREB and phospho-CREB were expressed in ANSCs with or without application of propofol. Propofol up-regulated the phosphorylation level of CREB in ANSCs. Conclusion Propofol promotes the proliferation of ANSCs in vitro via up-regulation of phosphorylation level of CREB in ANSCs.
