丙泊酚与咪达唑仑对离体细胞内毒素损伤后炎性反应的影响

摘要:

目的 探讨丙泊酚与咪达唑仑对离体细胞内毒素损伤后炎性反应的影响。方法 将内毒素诱导血清分为A、B、C、D四组:A组:对照组;B组:丙泊酚+咪达唑仑组,加入丙泊酚和咪唑安定配制成含有两药10μmol/L;C组:咪唑安定组含咪达唑仑10 μmol/L;D组:丙泊酚组含丙泊酚10 μmol/L。分别在给药前(T0)、给药后即刻(T1)、80 min(T2)、120 min(T3)、180 min(T4)时间点采用化学发光法进行测定肿瘤坏死因子α(TNF-α)的浓度。结果 T0-T2 时,各组间血清TNF 活性差异无统计学意义,T3-T4 时各试验组 TNF 活性均明显低于对照组(3121.0±157.8 和 1720.0±902.0),各试验组之间差异无统计学意义。结论 丙泊酚与咪达唑仑能较好的抑制离体细胞内毒素损伤后炎性因子的释放。

Abstract:

Objective To explore the effects of propofol and midazolam on intracellular toxin-induced inflammatory reaction in isolated cells. Methods Monocytes were collected from rabbit blood and cultured in media containing placebo (group A), 10 μmol/L of midazolam (group C), 10 μmol/L of propofol (group D) or 10 μmol/L of both midazolam and propofol (group B). Endotoxin LPS was delivered into the media later. TNF-α level was detected with chemiluminescent method before and immediately after, 80, 120, 180 min after LPS delivery, respectively. Results Both propofol and midazolam significantly reduced the level of TNF-α released by isolated monocytes in the presence of LPS. Conclusion Propofol and midazolam can inhibit the release of intracellular toxin-induced inflammatory factors.
