Abstract: The major limitation to a long-term use of opioids, such as morphine, is the development of physiological tolerance, observed as a profound decrease in analgesic effect. Although the molecular and cellular mechanisms underlying the development of morphine tolerance remain controversial, great progress has been made. This review focuses on the opioid receptor with the aim of gaining a greater insight into the mechanism of morphine tolerance to analgesia.
