线粒体钙单向转运体在二氮嗪预处理脑保护中的作用

摘要:

目的 探讨线粒体钙单向转运体是否参与了二氮嗪预处理所发挥的脑保护作用 方法 将32只健康雄性Wistar大鼠随机分为4组:A组,假手术组;B组,脑缺血再灌注组;C组,脑缺血再灌注+二氮嗪组;D组,脑缺血再灌注+二氮嗪+精胺组。采用线栓法建立大鼠大脑中动脉闭塞(MCAO)模型,缺血2h再灌注24h后观测各组神经行为变化,脑组织SOD活性、MDA含量、NO含量和GSH-PX活性。结果 与B组相比,C组二氮嗪预处理可以改善大鼠脑缺血再灌注引起的神经行为障碍(P<0.01),提高SOD及GSH-PX活性、降低MDA及NO含量(P<0.05)。与C组相比,D组精胺可减弱二氮嗪预处理改善神经行为障碍(P<0.01)、提高SOD和GSH-Px活性以及降低MDA和NO含量的效应(P<0.05)。结论 二氮嗪预处理通过抑制过氧化损伤诱导的抗大鼠脑缺血再灌注的保护作用,可能与再灌注时线粒体钙单向转运体受到抑制有关。

Abstract:

Objective To investigate whether diazoxide preconditioning protects cerebral against brain ischemia reperfusion via mitochondrial calcium uniporter or not.Methods The ischemia reperfusion models were performed by thread occlusion of middle cerebral artery for 2h,and then 24h reperfusion.32 rats were randomly divided into four groups:A group(sham-operated group), B group(ischemia reperfusion group), C group(diazoxide group), D group(diazozide-spermine group).After 24h reperfusion,neuropsychological score,the activity of superoxide dismutase(SOD) and glutathione peroxidase(GSH-Px),the content of malondialdehyde(MDA) and nitric oxide(NO) were observed in above groups respectively.Results Compared with B group,diazoxide preconditioning of C group improved neuropsychological score(P<0.01),improved the activity of SOD and GSH-Px(P<0.05),and decreased the content of MDA and NO(P<0.05). Compared with C group,spermine attenuated the effects of diazoxide preconditioning of C group, which improved neuropsychological score(P<0.01),improved the activity of SOD and GSH-Px(P<0.05),and decreased the content of MDA and NO(P<0.05).Conclusions The effects of diazoxide preconditioning against brain ischemia reperfusion by attenuating peroxidation injury in rats may involve in the suppression of MCU during reperfusion.
