茶多酚对大鼠全脑缺血/再灌注损伤后神经元凋亡和神经行为学的影响

摘要:

目的:观察茶多酚对大鼠全脑缺血/再灌注(ischemia/reperfusion, I/R)损伤后海马CA1区神经元凋亡和神经行为学改变的影响。方法:健康清洁雄性SD大鼠216只,按随机数字表法分为3组(每组72只),分别为假手术组(shan operation, SO组),缺血/再灌注模型组(I/R组)和茶多酚组(tea polyphenols,TP组,TP 200mg/kg再灌注即刻腹腔注射),采用DNA断裂的原位末端标记法(TUNEL法)以及流式细胞仪检测细胞凋亡,利用旷场实验和高架十字迷宫实验评价神经行为学变化。结果:大鼠全脑I/R后,TUNEL法结果光镜下观察显示:I/R组海马CA1区可见较多凋亡细胞,TP组凋亡细胞数较I/R组明显减少,流式细胞仪检测结果显示:全脑I/R后48 h,TP组细胞凋亡率达18.77%,I/R组细胞凋亡率达26.47%,TP组与I/R组相比明显降低,两组间差别具有统计学意义(P<0.05)。旷场试验和高架十字迷宫实验结果显示:与I/R组相比,TP组大鼠探索能力增强,焦虑情绪减轻,差别具有统计学意义(P<0.05)。结论:大鼠全脑I/R后,茶多酚能够发挥抗凋亡作用,降低海马CA1区神经元的凋亡率,并在抑制神经元凋亡基础上改善大鼠全脑I/R后的神经行为学功能。

Abstract:

Observe The influence of tea polyphenols on hippocampal neuronal apoptosis in CA1 region of rats after global cerebral ischemia/reperfusion(I/R) injury, and disscuss the relationship between rat’s neuronal behavior and hippocampal neuronal apoptosis in addition. Methods: 216 healthy male SD rats were randomly divided into 3 groups: sham operation group (SO group, n = 72), ischemia- reperfusion group (IR group, n = 72) and tea polyphenols group (TP group, n = 72, TP200mg/kg, TP were intraperitoneally injected at the time of reperfision), TUNEL was used to observe, qualitative and locate apoptotic neurons, flow cytometry was used to detected apoptotic index. The open field test and the high cross maze were used to test the rats’ neuroethology. Results: After cerebral ischemia / reperfusion injury, TUNEL assay showed that: compared with TP group, hippocampal neurons in CA1 region of I/R group can be seen more apoptotic cells. Apoptotic cells tested by flow cytometry showed: the apoptosis of TP group reached 26%, compared with the I/R group, the rate of apopotosis was significantly reduced (P <0.05).Open-field test and elevated plus maze experiments showed that: compared with I/R group, TP group showed some activity and the ability to explore and their emotions are more stable (P<0.05). Conclusions: After cerebral ischemia / reperfusion, tea polyphenols can play the role of anti-apoptosis, reducing the rate of cellular apoptosis in hippocampal CA1 region and it was able to improve rats’ neurological ethology based on this function.
