异氟醚和七氟醚对人肺腺癌A549细胞迁移及Cdc42、Fascin表达的影响

摘要:

目的 观察异氟醚和七氟醚对人肺腺癌A549细胞迁移及细胞分裂周期蛋白42(cell division cycle 42,Cdc42)和Fascin表达的影响。方法 人肺腺癌A549细胞接种于培养板,培养24 h后,随机分为对照组(C组)、2.3%异氟醚组(Iso组) 、3.4%七氟醚组(Sev组) 。Iso组和Sev 组的A549细胞分别暴露于2.3%异氟醚或3.4%七氟醚2、4、6 h。C组不接受异氟醚或七氟醚处理。三组处理结束后,继续培养24 h。于24 h时,采用Transwell法检测各组A549细胞处理2、4、6 h后的迁移细胞数目,蛋白印迹法检测各组A549细胞处理4 h的Cdc42和Fascin表达水平。结果 与C组和Iso组比较,Sev组的迁移细胞数目降低(P <0.05);与C组和Iso组比较,Sev组的Cdc42表达下调[(75±16)%,(79±13) % vs (31±7) %](P <0.05);与C组和Iso组比较,Sev组的Fascin表达下调[(71±13)%,(77±18)% vs (40±8)%](P <0.05)。结论 七氟醚能抑制人肺腺癌A549细胞的迁移,该效应与抑制Cdc42和Fascin表达有关。异氟醚无抑制A549细胞迁移的效应。

Abstract:

Objective To investigate the effects of isoflurane and sevoflurane on migration and expression of cell division cycle 42 (Cdc42), Fascin of human adenocarcinoma cell line A549. Methods A549 cells were inoculated in culture plate. After being cultured for 24 h the cells were randomly divided into 3 groups: group control (C), group 2.3 % isoflurane (Iso), and group 3.4% sevoflurane (Sev). Each group contained 6 wells. A549 cells of group Iso and Sev were exposed to 2.3% isoflurane or 3.4%sevoflurane for 2, 4, and 6 h respectively. Cells of group C were exposed to 95%O2-5%CO2 mixture air, and were then cultured for additional 24 h. The migration of A549 cells were assessed by Transwell cell culture chamber. The expression Cdc42 and Fascin in A549 cells were detected by Wester blotting at 24 h after isoflurane and sevoflurane treatment for 4 h. Results Compared with group C and group Sev,the number of migration cells in group Sev was significantly decreased (P <0.05). The expression of Cdc42 in group Sev was significantly down-regulated as compared with group C and group Iso [(75±16)%,(79±13) % vs (31±7) %] (P <0.05); The expression of Fascin in group Sev was significantly down-regulated as compared with group C and group Iso [(71±13)%,(77±18)% vs (40±8)%](P <0.05). Conclusion Sevoflurane can inhibit the migration of A549 cells through down-regulating the expression of Cdc42 and Fascin. Isoflurane can not inhibit the migration of A549 cells.
