地佐辛在小鼠神经病理性疼痛-瘙痒共病中的治疗效果

Therapeutic Efficacy of Dezocine for the Treatment of the Co-morbidity of Neuropathic Pain and Itching in Mice.

摘要:

目的 观察地佐辛在小鼠坐骨神经分支高选择结扎切断 (spared nerve injury, SNI)诱导的神经病理性疼痛与氯喹激发的急性瘙痒共病(co-morbidity of pain-itch, CPI)中的治疗效果。方法 成年雄性SPF级昆明小鼠,体重(22-26) g,随机分为6组(n=8):A组,假手术-非瘙痒-生理盐水治疗对照组(Sham+NS+NS);B组,神经病理性疼痛-非瘙痒-生理盐水治疗对照组(SNI+NS+NS);C组,神经病理性疼痛-瘙痒-生理盐水治疗对照组(SNI+Itch+NS);D组,假手术-瘙痒-生理盐水治疗对照组(Sham+Itch+NS);E组,神经病理性疼痛-瘙痒-地佐辛治疗组(SNI+Itch+Dezocine);F组,假手术-瘙痒-地佐辛治疗组(Sham+Itch+ Dezocine)。于SNI术前1d,术后1d, 3d, 5d(地佐辛/氯喹注药前、后),采用电子VonFrey测定各时间点小鼠机械缩足阈值(pal withdrawal threshold, PWT)评估机械痛敏;采用视频记录并分析动物因氯喹激发的搔抓行为。结果 各组机械痛敏/镇痛效应PWT曲线下面积(AUC)比较:F-AUC>E-AUC>B-AUC;D-AUC>C-AUC (P<0.05)。搔抓潜伏期(scratching latency, SL)比较:A组(49±11)秒,B组(51±10)秒,C组(322±127),D组(333±94)秒,E组(571±67)秒,F组(524±77)秒;地佐辛治疗延长小鼠的搔抓潜伏期(P<0.05)。搔抓次数比较:C组(13.67±3.39),D组(25.78±5.21),E组(5.78±1.99),F组(6.33±1.80),C组搔抓次数< D组搔抓次数(P<0.05),E、F组搔抓次数< C、D组搔抓次数(P<0.05)。结论 对于小鼠CPI的治疗,地佐辛具有显著的镇痛与抗瘙痒作用。

Abstract:

Objective To observe the therapeutic Efficacy of dezocine on the treatment for the co-morbidity of neuropathic pain and itching, which induced by spared nerve injury (SNI) and evoked by chloroquine intradermal injection. Methods Adult male Kun-Ming mice (SPF grade, 22–26-g), were assigned randomly to six groups (n = 8): Group A (Sham+NS+NS), Group B (SNI+NS+NS), Group C (SNI+Itch+NS), Group D (Sham+Itch+NS), Group E (SNI+Itch+Dezocine), and Group F (Sham+Itch+ Dezocine). Mechanical allodynia was assessed by using electron-Von Frey. To test pain threshold, Paw withdrawal threshold (PWT, to assess mechanical allodynia) was measured by ElectroVonFrey (IITC Life Science, USA) on -1d (baseline), 1 d, 3 d, and 5 d (before and after dezocine and chloroquine intradermal injection). Scratching behaviors of animals were video-recorded to analysis chloroquine-evoked itching in each group.
