肺缺血/再灌注对大鼠肺热休克蛋白70表达的影响

Effects of lung ischemia/reperfusion on heat shock protein 70 expression in the lung of rats

摘要:

目的 探讨肺缺血/再灌注不同时间对大鼠肺泡灌洗液(bronchoalveolar lavage fluid, BALF)中的热休克蛋白(heat shock protein, HSP)70表达的影响。 方法 将50只雄性SD大鼠按完全随机分组方法分成假手术组(Sham组)、缺血组(I组)、缺血/再灌注30 min组(IR1组)、缺血/再灌注60 min组(IR2组)和缺血/再灌注90 min组(IR3组),每组10只。用无创动脉夹夹闭肺门方式复制大鼠原位肺缺血/再灌注模型。分别测定大鼠BALF中超氧化物歧化酶(superoxide dismutase, SOD)活性和HSP70含量。 结果 I组BALF中SOD含量和HSP70浓度分别为(11.00±0.51) ×103 U/L和(93±5) ng/L,与Sham组比较明显升高,差异有统计学意义(P<0.05);随再灌注时间的延长,SOD含量在IR1组[(9.97±0.80) ×103 U/L]、IR2组[(9.07±0.40) ×103 U/L]和IR3组[(8.08±0.26) ×103 U/L]中逐渐降低,差异有统计学意义(P<0.05);而HSP70含量在IR1组[(103±6) ng/L]、IR2组[(116±10) ng/L]和IR3组[(136±6) ng/L]中逐渐增高,差异有统计学意义(P<0.05)。 结论 在再灌注90 min内,大鼠HSP70表达增强,并通过介导各种反应对机体损伤起保护作用。

Abstract:

Objective To discuss the influence of different durations of ischemia/reperfusion on the expression of heat shock protein (HSP) 70 in bronchoalveolar lavage fluid (BALF) in rats. Methods Fifty male SD rats were randomly divided into five groups by random number table method (n=10): Sham group, the left pulmonary hilum was dissociated after thoracotomy without occlusion and mechanically ventilated to the end. Ischemia group(I group), the left pulmonary hilum was clamped for 60 min. Ischemia/reperfusion with 30 min group(IR1 group), ischemia/reperfusion with 60 min group(IR2 group),Ischemia/reperfusion with 90 min group(IR3 group). Single lung ischemia and reperfusion animal model were developed by clamping the left pulmonary hilum noninvasive arterial clip. After the operation samples of lung tissues and BALF of all the rats in each group were analyzed. Results HSP70 concentration and superoxide dismutase(SOD) level form bronchoalveolar lavage fluid in I group were (11.00±0.51) ×103 U/L and (93±5) ng/L, significantly higher (P<0.05) than those in Sham group. As the extension of the duration of reperfusion, the SOD level in IR1 group (9.97±0.80) ×103 U/L, IR2 group (9.07±0.40) ×103 U/L and IR3 group (8.08±0.26) ×103 U/L, IR groups decreased gradually (P<0.05), while the content of HSP70 in IR1 group (103±6) ng/L, IR2 group (116±10) ng/L and IR3 group (136±6) ng/L gradually increased(P<0.05). Conclusions Reperfusion within 90 min increases the expression of HSP70, it may protect the lung against ischemia/reperfusion injury via a variety of reactions.
