参麦注射液对大鼠内毒素性急性肺损伤时JAK2/STAT3通路的影响

Effect of Shen-mai injection on janus kinase 2/signal transducer and activator of transcription 3 pathway in rats with lipopolysaccharide-induced lung injury

【摘要】 目的 探讨参麦注射液对大鼠内毒素性急性肺损伤时蛋白酪氨酸激酶2(janus kinase 2, JAK2)/信号转导和转录激活因子3(signal transducer and activator of transcription 3, STAT3)通路的影响。方法 健康成年雄性Wister大鼠采用随机数字表法分为3组:正常对照组(C组)、内毒素性肺损伤组(ALI组)和内毒素性肺损伤+参麦注射液处理组(SM组)。ALI组和SM组腹腔注射LPS 5 mg/kg,C组给予等容量生理盐水。SM组在LPS注射5 min后腹腔注射参麦注射液2 ml/只,C组和ALI组同时给予等容量生理盐水。收集支气管肺泡灌洗液(bronchoalveolar lavage fluid,BALF),测BALF中白细胞介素-1β(interleukin-1β,IL-1β)、肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)及细胞间黏附分子-1(intercellular adhesion molecule -1,ICAM-1)的浓度;取肺组织,测湿干重比(wet/dry,W/D比)、肺组织IL-1β、TNF-α和ICAM-1 mRNA的表达、采用免疫组化法检测肺组织p-JAK2/p-STAT3的表达水平、观察病理学结果并进行肺损伤评分。结果 SM组W/D比、肺损伤评分分别为(6.4±2.1)、(7.2±2.9),较ALI组(8.2±2.6)、(12.1±3.2)显著降低(P<0.05);SM组BALF中IL-1β、TNF-α及ICAM-1浓度分别为(53±10)、(94±19)、(187±36)ng/L,较ALI组(71±11)、(141±27)、(273±41)ng/L显著降低(P<0.05);SM组肺组织IL-1β、TNF-α、ICAM-1 mRNA含量分别为(1.71±0.06)、(1.81±0.07)、(1.61±0.08),较ALI组(2.64±0.14)、(2.71±0.10)、(2.59±0.09)显著降低(P<0.05);SM组肺组织p-JAK2和p-STAT3表达量分别为(0.33±0.09)、(0.32±0.09),较ALI组(0.45±0.03)、(0.45±0.04)显著降低(P<0.05)。结论 参麦注射液可能通过抑制JAK2/STAT3通路的激活减轻大鼠内毒素性急性肺损伤。

Abstract:

[Abstract] Objective To evaluate the effect of Shen-mai injection on janus kinase 2/signal transducer and activator of transcription 3 (JAK2/STAT3) pathway in mice with lipopolysaccharide-induced lung injury. Methods Thirty adult male Wister rats were randomly divided into 3 groups :control group(group C), LPS-induced lung injury group(group ALI), LPS-induced lung injury + Shen-mai injection group(group SM). LPS 5 mg/kg was injected intraperitoneally in group ALI and group SM, and then Shen-mai injection 2 ml was injected intraperitoneally. The left lung was lavaged, and broncho-alveolar lavage fluid(BALF)was collected for determination of concentrations of protein,tumor necrosis factor-α (TNF-α), interleukin-1β(IL-1β) and intercellular adhesion molecule-1(ICAM-1). The lung tissues were removed for determination of the wet to dry lung weight(W/D) ratio, the mRNA expression level of IL-1β, TNF-α and ICAM-1. The pathological changes of the lungs were determinated by using light microscope and the lung injury scores were also determinated and immunohistochemistry was used to detected the protein expression level of p-JAK2, p-STAT3,. Results The W/D ratio and lung scores were significantly decreased in group SM (6.4±2.1),(7.2±2.9),compared with group ALI(8.2±2.6)、(12.1±3.2). The concentrations of IL-1β, TNF-α and ICAM-1 in BALF(53±10)、(94±19)、(187±36)ng/L were significantly decreased than that in group ALI (71±11)、(141±27)、(273±41)ng/L. The protein expression level of p-JAK2 and p-STAT3 (0.33±0.09)、(0.32±0.09) were significantly decreased than that in group ALI (0.45±0.03)、(0.45±0.05). Conclusion Shen-mai injection attenuates lipopolysaccharide-induced lung injury in rats possibly through inhibiting JAK2/STAT3 pathway.
