Abstract: Background T cell death associated gene 8(TDAG8) is a proton sensing G protein coupled receptor which was widely expressed in the body. However, its biological function and clinical relevance still remain unknown. With the advances of research on TDAG8, especially the discovery of allosteric agonists and negative allosteric modulators of TDAG8 which provided important tools for further research, the physiological function and role of TDAG8 were gradually exposed. Objective This review focuses on several functions may regulated by TDAG8 and demonstrates biological function and clinical relevance of TDAG8. This summary provides important theoretical basis for further research. Content Systematically introduce TDAG8 physiological function and mechanism. Trend TDAG8 inflammation may be a critical mediator of inflammation.
