二丙烯三硫化物对大鼠急性肺损伤的保护作用

Protective effect of diallyl trisulfide on acute lung injury in rats

摘要:

目的 观察二丙烯三硫化物(diallyl trisulfide, DATS)对盲肠结扎穿孔(cecal ligation and puncture, CLP)诱导大鼠急性肺损伤的保护作用。 方法 采用CLP法制作急性肺损伤动物模型。30只雄性SD大鼠术前禁食16 h,腹腔注射10%水合氯醛4 ml/kg麻醉后,采用随机数字表法将动物分成3组(每组10只): sham手术组(Ⅰ组)、CLP+生理盐水(normal saline, NS)组(Ⅱ组)和CLP+DATS组(Ⅲ组)。Ⅰ组和Ⅱ组经尾静脉注射NS 10 ml/kg,而Ⅲ组尾静脉注射DATS 10 mg/kg。模型制作完毕后0 h(T0)、6 h(T6) 和12 h(T12)经大鼠左颈总动脉抽动脉血测动脉血气。术后所有30只雄性SD大鼠皮下注射生理盐水0.05 ml/g。手术后12 h处死SD大鼠留取肺组织标本,测定肺组织湿/干重比(wet/dry weight ratio, W/D)、髓过氧化物酶(myeloperoxidase, MPO)活性、超氧化物歧化酶(superoxide dismutase, SOD)活性、丙二醛(malondialdehyde, MDA)和一氧化氮(nitric oxide, NO)含量,测定肺组织胞质NF-κB P65、NF-κB抑制蛋白α(inhibitor of NF-κB, IκB-α)和胞核NF-κB P65的水平,光镜下观察大鼠肺组织的病理变化。另取30只雄性SD大鼠,分组和实验方法同前,观察3组大鼠24 h生存率。 结果 与Ⅰ组比较,Ⅱ组和Ⅲ组T6和T12时PaO2及SOD活性降低(P<0.05),PaCO2、肺组织W/D、MPO活性、MDA和NO含量升高(P<0.05)。与Ⅰ组比较,Ⅱ组光镜下炎症反应明显加重,胞质NF-κB P65和IκB-α减少(P<0.05),胞核NF-κB P65增加(P<0.05),24 h生存率降低(P<0.05)。与 Ⅱ组比较,Ⅲ组T6和T12时 PaO2、SOD活性增加(P<0.05),PaCO2、肺组织W/D、MPO活性、MDA和NO含量降低(P<0.05),光镜下肺组织的炎症反应明显减轻,胞质NF?蛳κB P65和IκB-α增加(P<0.05),胞核NF-κB P65减少(P<0.05),24 h生存率提高(P<0.05)。 结论 DATS对CLP诱导的SD大鼠急性肺损伤有保护作用。

Abstract:

Objective Observing the protective effect of diallyl trisulfide (DATS) on acute lung injury induced by cecal ligation and puncture (CLP). Methods The animal model of acute lung injury was induced by CLP. The 30 male SD rats were fasted 16 h before surgery, then was anesthetized by intraperitoneal injection of 10% chloral hydrate (4 ml/kg). Then all rats were randomly divided into 3 groups (n=10), sham group (group Ⅰ), CLP+normal saline (NS) group (groupⅡ) and CLP+DATS group (group Ⅲ). Rats in groupⅠ and group Ⅱ were injected with saline 10 ml/kg through caudal vein, but group Ⅲ were injected with DATS 10 mg/kg. The arterial cannulation was used in carotid artery of the rat to measure the blood gas after the surgery at 0 h (T0), 6 h (T6) and 12 h (T12). All male SD rats were subcutaneously injected with normal saline 0.05 ml/g. Subsequently, the male SD rats were sacrificed at 12 h after operation by the blooding and collected samples of lung tissue. The lung tissue wet/dry weight ratio (W/D), myeloperoxidase (MPO) activity, malondialdehyde (MDA), superoxide dismutase (SOD) and nitric oxide (NO) levels, expression of nuclear factor-κB(NF-κB) P65 and IκB-α in the cytoplasm of lung tissue, expression of NF-κB P65 in the nucleus of lung tissue were measured. Pathological changes of lung tissue was observed under light microscope. Another 30 male SD rats were grouped randomly and received the same experiment methods. The survival rate at 24 h was observed. Results Compared with group Ⅰ, the arterial partial pressure of oxygen (PaO2) and activity of SOD were significantly decreased (P<0.05), but the arterial partial pressure of carbon dioxide (PaCO2), lung tissue W/D, activity of MPO, concentration of MDA and NO in the group Ⅱ and group Ⅲ at T6 and T12 were significantly increased (P<0.05). Inflammatory reaction was significantly increased under light microscope in the group Ⅱ. The expression of cytoplasmic NF-κB P65 and IκB-α was decreased (P<0.05), but the expression of nucleus NF-κB P65 was increased in the group Ⅱ(P<0.05). The survival rate of the group Ⅱ in 24 h was lower than the rate in groupⅠ(P<0.05). Compared with groupⅡ, the arterial PaO2 and activity of SOD was increased (P<0.05), but the arterial PaCO2, lung tissue W/D, activity of MPO, concentration of MDA and NO were decreased in the group Ⅲ at T6 and T12(P<0.05). Inflammatory reaction was significantly decreased under light microscope in group Ⅲ. The expression of cytoplasmic NF-κB P65 and IκB-α were increased (P<0.05), but the expression of nucleus NF-κB P65 were decreased in group Ⅲ (P<0.05). The survival rate of the group Ⅲ in 24 h was improved (P<0.05). Conclusions DATS has a protective effect on acute lung injury in SD rats.
