摘要:

背景 Semaphorin 3A(Sema3A)是一种蛋白,最初被认为是抑制可轴突生长的物质。随后发现Sema3A在健康的椎间盘细胞中高表达,在退化样本中表达显著降低。 目的 讨论椎间盘源性疼痛是否与Sema3A表达有关,及Sema3A在疼痛中的发展作用。 内容 Semaphorins即信号素家族其结构由保守的500个氨基在末端组成七叶片b螺旋桨信号素结构域。该信号素家族有两个主要受体,分别是丛蛋白家族和神经纤维蛋白(neuropilin, Nrp)。Sema3A参与抑制轴突的生长,并参与机体骨量调节,与神经性疼痛的调节和椎间盘退变(intervertebral disc degeneration, IVD)的发展有关。 趋向 Sema3A可能可以作为调控椎间盘源性疼痛的新靶点并可能具有诊断和治疗椎间疾病的潜力。

Abstract:

Background Semaphorin 3A(Sema3A) is a protein identified originally as a diffusible axonal chemorepellent. There is no nerve innervation into healthy disc cells whereas nerve grows into diseased intervertebral disc in chronic back pain. Objective To discuss if Sema3A is related to discogenic pain and the role of Sema3A in the development of discogenic pain. Content The semaphorin family has a conserved 500 amino-terminalseven-bladed b-propeller semaphorin domain. The two major receptors of semaphorins are plexins family and neuropilins (Nrp1) and Nrp2. Sema3A can inhibit the outgrowth of axon and is correlated with the regulation of bone mass, neuropathic pain and the development of intervertebral disc degeneration (IVD). Trend Sema3A may be a new target in the regulation of discogenic pain. It is also a potential target in the diagnosis and therapy of intervertebral diseases
