丙泊酚麻醉对新生鼠海马区B淋巴细胞瘤-2蛋白及其相关蛋白表达的影响

摘要:

目的 探讨丙泊酚麻醉对新生鼠海马区B淋巴细胞瘤-2蛋白(B-cell lymphoma-2, Bcl-2)及其相关蛋白(Bcl-2 associated protein, Bax)表达的影响。 方法 新生7 d SD 大鼠112只,按随机数字表法分为8组(每组14只):对照组1(Con1组),腹腔连续注射5 d生理盐水;P50 组、P100组、P150组,分别连续5 d腹腔注射50、100 mg/kg和150 mg/kg丙泊酚;对照组2(Con2组),腹腔连续注射5 d生理盐水,最后1 d在丙泊酚麻醉下行阑尾切除术;SP50组、SP100组和SP150组,分别连续5 d腹腔注射50、100 mg/kg和150 mg/kg丙泊酚,最后1 d在丙泊酚麻醉下行阑尾切除术。实验结束饲养2个月后,取大鼠脑第5段的海马结构, 免疫组化法测定海马Bcl-2(抑制凋亡蛋白)、Bax(促凋亡蛋白)的表达。 结果 与Con1组比较,P50组、P100组、P150组Bax表达逐渐升高(P<0.05),Bcl-2表达逐渐降低,P100组、P150组差异有统计学意义(P<0.05);与P50组比较,P100组、P150组Bax表达升高(P<0.05),Bcl-2表达降低(P<0.05);与Con2组比较,SP50组、SP100组、SP150组Bax表达逐渐升高(P<0.05),SP50组、SP150组Bcl-2表达逐渐降低(P<0.05);与SP50组比较,SP100组、SP150组Bax表达升高(P<0.05),SP150组Bcl-2表达降低(P<0.05)。 结论 反复丙泊酚麻醉通过抑制Bcl-2的活化,增强Bax的表达,可促进新生大鼠海马神经元凋亡,且呈剂量相关性。

Abstract:

Objective To investigate the effects of propofol on the expression of B-cell lymphoma-2 assaciated protein (Bax) and B-cell lymphoma-2(Bcl-2) in the hippocampus of neonatal rats. Methods A total of 112 male newborn Sprague Dawley (SD) rats aging 7 d were randomly divided into 8 groups (n=14): control group (Con1 group, intraperitoneally given normal saline for 5 d), and P50, P100 and P150 groups which received 50, 100 mg/kg and 150 mg/kg propofol once per day for 5 d, respectively; and control 2 group (Con2 group, intraperitoneallyl receiving normal saline for 5 d, and appendectomy was performed under propofol anesthesia for the last day) and SP50, SP100 and SP150 groups which received 50, 100 mg/kg and 150 mg/kg propofol for 5 d respectively, and appendectomy was performed under propofol anesthesia for the last day). Two months after the experiments, the level of Bcl-2 (inhibitor of apoptosis proteins) and Bax (the proapoptotic protein) were detected using immunohistochemical methods. Results Compared with the Con1 group, the levels of Bax in the P50, P100 and P150 groups increased, while the amounts of Bcl-2 gradually decreased in the P100 and P150 groups(P<0.05). Compared with the P50 group, the levels of Bax increased (P<0.05), while the amounts of Bcl-2 decreased in the P100 and P150 groups. Compared with the Con2 group, the level of Bax in the SP50, SP100 and SP150 groups gradually increased (P<0.05), while the amount of Bcl-2 in the SP50 and SP150 groups gradually decreased (P<0.05). Compared with the SP50 group, the level of Bax in the SP100 and SP150 groups increased (P<0.05), while the amounts of Bcl-2 was decreased in the SP150 group (P<0.05). Conclusions Repeated administration of propofol can inhibit the activation of Bcl-2 and enhance the expression of Bax. It can promote the apoptosis of hippocampal neurons of newborn rats in a dose-dependent manor.
