Abstract: Programmed cell death mainly includes pyroptosis, apoptosis, necroptosis, and other pathways, which play an important role in host defense against pathogen invasion and maintaining homeostasis. Although previous studies have believed that various programmed cell death pathways are independent pathways. Recently increasing evidence pointed to complex crosstalk between these signaling pathways. This process is termed PANoptosis, and it is regulated by upstream PANoptosome. In this review, we aim to provide a broader perspective for the prevention and treatment of acute lung injury (ALI)/acute respiratory distress syndrome (ARDS) by describing the constituent sources, stimuli, mechanisms of formation and progress of PANoptosis and PANoptosome complex.
