鞘内注射Akt抑制剂Ⅳ对注射式根性神经痛大鼠痛行为学的影响

Effects of intrathecal injection of Akt inhibitor Ⅳ on pain behavior in rats model of injected radicular pain

摘要:

目的 探讨鞘内给予Akt抑制剂Ⅳ对注射式根性神经痛大鼠痛行为学的影响。方法 选择成功鞘内置管后无运动障碍的雄性SD大鼠18只,随机分为假手术组(S组)、溶剂对照组(C组)和 Akt抑制剂 组(A组),每组6只。C 组和A 组均采用注射surgiflo制备注射式根性神经痛模型, A 组于造模前 30min 给予Akt抑制剂Ⅳ 0.6 μg/10 μl。S 组和 C 组则给予等量的 10% 酒精。各组均于造模前、造模后第1 d, 4 d, 7 d, 10 d, 17 d, 28 d 测量双侧足底机械缩足阈值(Paw withdrawal mechanical threshold, PWMT)和热辐射刺激潜伏期(Paw withdrawal thermal latency, PWTL)(n=6)。结果 与 S 组相比,造模后第1 d C 组同侧肢体 PWMT(9.56±1.49 vs 14.03±0.94)和 PWTL (9.91±1.80 vs 19.96±1.69)明显降低(p<0.05), 持续至术后28 d ;而对侧PWMT和 PWTL与 S 组则无明显变化 (p> 0.05)。与 C 组相比, A 组痛行为学明显增加(PWMT:4.88±1.36 vs 7.51±0.88;PWTL:5.83±1.19 vs 8.19±1.66),并持续至术后7 d。结论 鞘内注射Akt抑制剂Ⅳ可以加重慢性背根神经节 压迫引起的痛行为学,Akt在注射式根性神经痛大鼠痛行为学的形成中起重要作用。

Abstract:

Objective To investigate the effects of intrathecal injection of Akt inhibitor Ⅳ on pain behavior in rats model of injected radicular pain. Methods Eighteen male Sprague Dawley(SD) rats with succeed intrathecal intubation were randomly divided into three groups(n=6): Sham group(S), injected radicular pain group(C), Akt inhibitor group(A). The C group and A group of injected radicular pain were established by surgiflo injection. Akt inhibitorⅣ was injected at 30min preoperative day in group A.Vehicle(10% ethanol) was intrathecally injected at the same time in group C and group S. Pain ethology indexes such as paw withdrawal mechanical threshold (PWMT) and paw withdrawal thermal latency (PWTL) were assessed on 1d before and 1 d , 4 d, 7 d, 10 d, 17d and 28d after operation. Results Compared to group S and preoperative level, PWMT and PWTL were decreased at 1 postoperative day in group C (p<0.05). Intrathecal Akt inhibitorⅣ injection significantly decreased PWMT and PWTL (p<0.05) in groups A . Conclusion Intrathecal Akt inhibitorⅣ injection would increase pain behavior in rat model of injected radicular pain. Akt would play an important role in the developing pain behavior in rat model of injected radicular pain.
