不同剂量布托啡诺硬膜外注射对大鼠血清MBP与脊髓形态学的影响

摘要:

目的 观察硬膜外注射不同剂量的布托啡诺对大鼠脊髓形态功能与血清髓鞘碱性蛋白(MBP)的影响。方法 雄性SD大鼠,体重180~210g,于L1~2处行硬膜外置入PE-530导管,3d后,取无运动障碍的大鼠32只,随机分为4组,每组8只:生理盐水组(NS组)、布托啡诺1、2、3组(B1组、B2组、B3组)。C组硬膜外注射NS 30μl,B1、B2、B3组硬膜外分别注射布托啡诺60、120和240μg(均用NS稀释至30μl)。置管成功后第1d~5d给药,1次/d。动物在给药前(T0)、末次给药后6h(T1)、24h(T2)、72h(T3)采血,检测血清MBP。末次给药后72h(T3),取脊髓腰膨大,行HE染色、Bcl-2、Bax、细胞凋亡和电镜观察。结果 各组血清MBP未见明显差异。和生理盐水组(C)和低剂量组(B1)相比,中剂量组(B2)和高剂量组(B3)神经细胞变性坏死,尼氏体消失;bcl-2表达明显减少,Bax表达明显增加,凋亡细胞明显增多 (P<0.05);髓鞘数量减少,结构破坏。结论 布托啡诺60μg连续给药对大鼠脊髓形态功能无影响,但布托啡诺120μg 和布托啡诺240μg连续给药会损害大鼠脊髓形态功能。

Abstract:

【Abstract】 Objective To observe the effect of epidural injection with different dose of butorphanol on the rats’ serum myelin basic protein(MBP) and spinal cord morphology. Methods A PE-530 catheter was inserted into the epidural space of each Sprague-Dawley rat (male,weighting 180-210g) at L1-2 level,and three days later,a total of 32 rats without any motor dysfunction were randomly divided into 4 groups as follow:saline group (group NS,n=8)and butorphanol group( B1,n=8;B2,n=8;B3,n=8).Rats in group NS were epidurally injected NS 30μl each ,and rats in group B1,B2 and B3 were respectively epidurally injected butorphanol 60μg,120ug and 240μg(all diluted to 30μl with NS)1 time per day 5 days, the serum MBP of rats were measured before injection (T0),6h(T1),24h(T2) and 72h (T3) after the last injection by ELISA examination.At T3,the lumbar enlargement of the spinal cord,was taken for detecting the morphology and apoptosis with optical microscope and electron microscope. Results There was no significant difference between each group in content of the serum MBP (P>0.05).In group B2 and B3,compared with group NS and B1,the neurons showed degeneration and necrosis,the nissl body disappeared and the expression of the Bcl-2 decreased obviously while the Bax increased.Besides that,in group B2 and B3,the number of apoptosis neurons increased obviously (P<0.05) and the number of myelins diminished,while the structure of myelins was destroyed. Conclusion Repeated epidural injection of butorphanol 60ug has no influence on spinal cord morphology of rats,while injection of 120ug and 240ug can impair the spinal cord morphology.
