摘要:

目的 探讨氨磷汀对谷氨酸所致N9小胶质细胞损伤的影响。 方法 用含1mM氨磷汀和10mM 谷氨酸的培养液培养N9细胞24h后,检测细胞代谢率、乳酸脱氢酶(LDH)释放量及培养液中超氧化物歧化酶(SOD)、一氧化氮(NO)和谷胱甘肽(GSH)的含量。 结果 与损伤组相比1mM氨磷汀保护组细胞代谢率升高(P<0.05),LDH释放减少(P<0.05),SOD、GSH含量增加(P<0.01),NO释放量下降(P<0.01);该作用可被兴奋性氨基酸转运体拮抗剂TBOA逆转。 结论 氨磷汀通过抗氧化机制减轻谷氨酸对N9小胶质细胞的损伤。

Abstract:

Objective To investigate the effects of Amifostine on glutamate induced microglia injury. Methods N9 microgial was treated with 1mM Amifostine and 10mM glutamate. Metabolism of microglial, LDH release, SOD activity, NO release and GSH level were evaluated. Results As compared to control group, microglial metabolism, SOD activity and GSH level were increased, while LDH and NO were decreased in Amifostine group. The effects of Amifostine could be reversed by the antagonist of excitatory amino acid transporters TBOA. Conclusion Amifostine protects N9 microglial glutamate injury through its antioxidantive effects.
