布比卡因致高糖培养SH-SY5Y细胞ROS增多与凋亡增加

【摘要】目的 探讨布比卡因作用于高糖培养的SH-SY5Y细胞内ROS增多和凋亡增加情况。方法 不同浓度葡萄糖培养基培养SHSY-5Y 细胞,经1mmol/L布比卡因作用6h。采用流式细胞仪检测细胞内ROS的产量及细胞凋亡状况、MTT法检测细胞存活率、Western blotting技术检测神经细胞内GRP78相关蛋白的变化。计量资料以均数±标准差(x ̅±s)表示,采用SPSS13.0统计软件分析。细胞存活率、ROS含量、细胞凋亡率和GRP78相关蛋白的变化采用单因素方差分析,组间比较采用LSD法(方差齐)或Dunnett's T3法(方差不齐)。P <0.05为差异有统计学意义。结果 各组含不同浓度糖培养基的细胞经布比卡因作用6h检测细胞内ROS含量。在葡萄糖浓度为5.56mmol/L、6.1 mmol/L、7 mmol/L组间ROS含量差异无统计学意义(P>0.05)。葡萄糖浓度为7.8 mmol/L、11.1mmol/L和13.3 mmol/L时,各组细胞内ROS含量差异有统计学意义(P< 0.01)。经布比卡因作用6h后,各组细胞相对存活率差异具有统计学意义(P<0.01)。组间差异均具统计学意义(P<0.05)。经布比卡因作用6h后各组细胞凋亡率差异具有统计学意义(P<0.01)。组间细胞凋亡率差异均具有统计学意义(P<0.01)。Western blot检测结果,单独效应分析发现在布比卡因处理6h后,5.56mmol/L组和6.1mmol/L组分别为1.02±0.12和0.97±0.06,均高于7.0mmol/L组(0.39±0.04),差异有统计学意义(P<0.01)。7.8mmol/L组(0.98±0.06)高于7.0mmol/L组(0.39±0.04),差异有统计学意义(P<0.01)。5.56mmol/L 组、 6.1mmol/L组和7.8mmol/L组相比,差异没有统计学意义(P>0.05)。11.1mmol/L组与其他各组相比,差异均具有统计学意义(P<0.05)。结论 细胞ROS增多及导致细胞凋亡增加与布比卡因进一步加强细胞内质网应激有关,引起细胞内ROS爆发,从而导致细胞损伤和凋亡。Grp78功能减弱导致细胞内质网应激进一步加强。

Abstract:

objective To discuss Bupivacaine act on SH-SY5Y cells in high sugar environment increasing ROS and apoptosis . Methods we adopt 1mmol/L bupivicaine to SH-SY5Y cells cultured by different concentrations of glucose medium ,and employ flow cytometry to detect the quantity of ROS in the cells and the situation of apoptosis.we use MTT method to detect cell survival rate .At last we use Western blotting technique to detect the change of the related protein about GRP78. Measurement data were presented as mean±standard deviation. SPSS 13.0 software was used for statistical treatment. The quantity of ROS and the situation of apotosis and cell survival rate and the quantity of GRP78 were analyzed by one-way ANOVA and comparisons among groups were made by LSD (homogeneous variance) or Dunnett’s T3 (heterogeneity of variance). P < 0.05 was considered statistically significant . Results The quantity of ROS in the cells cultured by glucose medium increasing gradually and the apoptosis and cell survival rate have statistical differences(P<0.01).Western blot results show that GRP78 express enhance in endoplasmic reticulum stress. Conclusion Intracellular ROS increasing and apoptosis are associated with Bupivacaine further strengthening endoplasmic reticulum stress. Endoplasmic reticulum stress cause ROS outbreak in cells, leading to cell damage and apoptosis. With the function of GRP78 degrading, endoplasmic reticulum stress further strengthen.
