脊髓CC趋化因子配体2在大鼠吗啡镇痛耐受中作用

摘要:

[摘要]目的 观察脊髓CC趋化因子配体2(CCL2)蛋白在大鼠吗啡镇痛耐受过程中的作用。方法 对SD大鼠连续7d鞘内注射吗啡(15μg)建立吗啡耐受的动物模型。采用50℃热水甩尾法观察CCL2在吗啡的镇痛耐受中作用;应用酶联免疫法(ELISA)检测吗啡耐受过程中脊髓背角CCL2免疫活性表达变化。结果 在吗啡注射3d﹑5d﹑7d,与IgG+吗啡组比较,CCL2 中和抗体4+吗啡组大鼠%MPE(最大镇痛效应百分率)明显增加(P<0.01),与IgG4+ 吗啡组比较,CCL2 中和抗体4+吗啡组大鼠%MPE明显增加(P<0.01);在吗啡注射9d﹑11d, 与IgG8+ 吗啡组比较,CCL2 中和抗体8+吗啡组大鼠%MPE明显增加(P<0.05,P<0.01); 在吗啡注射3d﹑5d﹑7d,与IgG + NS组比较, IgG +吗啡组大鼠脊髓背角CCL2的表达明显增加(P<0.05,P<0.01);然而,与IgG +吗啡组比较,CCL2 中和抗体+吗啡组大鼠脊髓背角CCL2的表达明显减低(P<0.05,P<0.01) 。结论 脊髓CCL2可能参与吗啡耐受形成和维持,抑制CCL2可能为临床吗啡镇痛耐受提供一种新的治疗。

Abstract:

[Abstract] Objective The present study was performed to investigate the possible role of spinal cord chemokine (C-C motif) ligand 2 (CCL2) in morphine antinociceptive tolerance. Methods Morphine antinociceptive tolerance was induced by intrathecal administration of 15 mg of morphine daily for 7 days. The role of CCL2 in morphine antinociceptive tolerance is explored by hot-water tail-flick test. ELISA(enzyme-linked immunosorbent assay )was used to test the change in spinal CCL2 immunoreactivity .Results On the 3rd ﹑5th and 7th day after chronic morphine , The %MPE (percent of maximal possible antinociceptive effect)were significantly decreaced in CCL2 neutralizine antibody plus morphine group as compared with IgG plus morphine group(P<0.05,P<0.01),The %MPE were significantly up-regulated in CCL2 neutralizine antibody plus morphine group as compared with IgG4 plus morphine group(P<0.01); On the9th and 11th day after chronic morphine,The %MPE were significantly up-regulated in CCL2 neutralizine antibody8 plus morphine group as compared with IgG8 plus morphine group(P<0.05,P<0.01); On the 3rd ﹑5th and 7th day after chronic morphine:CCL2 expression in IgG plus morphine group were significantly up-regulated as compared with IgG plus saline group(P<0.05,P<0.01), While the CCL2 expression in CCL2 neutralizine antibody plus morphine group were significantly down-regulated as compared with IgG plus saline group(P<0.05,P<0.01).Conclusions spinal CCL2 is involved in the development and maintenance of tolerance to morphine-induced analgesia. Inhibition of CCL2 may provide a new therapy for morphine tolerance management.
