依托咪酯乳剂对老年大鼠海马神经元凋亡及突触素表达的影响

摘要:

目的 观察腹腔注射依托咪酯乳剂对老年大鼠海马神经元突触素表达的影响。方法 选择月龄为18个月的SD大鼠45只,将所有大鼠随机分为3组(n=15),N组为生理盐水组,E组为依托咪酯组,L组为脂肪乳组。实验第1至第3天E组腹腔注射依托咪酯(乳剂)20mg/kg,,L组腹腔注射20%脂肪乳20mg/kg,N组腹腔注射同等体积的生理盐水,每天一次,连续3天。实验第4天,处死大鼠,取右侧脑海马组织,固定,石蜡切片,进行TUNEL检验、天冬氨酸特异性半胱氨酸蛋白酶(Caspase-3)蛋白电泳(westernblotting)及突触素免疫组化检验。结果 TUNEL结果:海马CA1区神经元凋亡,与N组、L组比较,E组海马神经元凋亡细胞及凋亡指数无明显增加,差别无统计学意义(P>0.05)。Caspase-3表达结果:海马CA1区神经元Caspase-3表达,与N组、L组比较,E组海马神经元活化Caspase-3片段表达无明显增加,差别无统计学意义(P>0.05)。三组大鼠的突触素结果:海马CA1区神经元突触素表达,与N组、L组比较,E组海马含突触素的神经元个数无明显减少,差别无统计学意义(P>0.05)。结论 腹腔注射依托咪酯乳剂20mg/kg对老年大鼠海马神经元的凋亡及突触素的表达无明显影响。

Abstract:

Objective To investigate the effects of etomidate emulsion on appotosis of hippocampua and changes of Synaptophysin in aged rats with instraperitoneal injection. Methods 45 SD rats of both sexes aged 18 months were divided into 3 groups: N: NS group (N, n=15),E: Etomidate group ( E, n=15); L: Lipid group (L, n=15). On the 1st day to 3rd day, the animals received intraperitoneal ( IP) with NS in the same volume ,or etomidate 20 mg/kg, or lipid 20 mg/kg once a day. On the 4th day, the animals were killed and the brains were immediately removed for determination of TUNEL , synaptophysin and caspase-3 in the hippocampua. Results TUNEL: The numbers of apoptosis cells in hippocampua were almost the same among 3 groups(P>0.05). Compared with group N and group L, AI of group E did not rise significantly(P>0.05). Synaptophysin: The expression of synaptophysin was the same among 3 groups, too(P>0.05). Caspase-3: The expression of activated Caspase-3 was the same among 3 groups, too(P>0.05). Conclusion Etomidate (20mg/kg) does not have any significant effect on apoptosis and changes of Synaptophysin in Hippocampal neuron of aged rats .
