红景天苷对机械通气致小鼠急性肺损伤的保护作用研究

Investigation of the role of salidroside in acute lung injury induced by mechanical ventilation in mice

【摘要 】 目的 探讨红景天苷(salidroside,SDS)对机械通气导致的小鼠急性肺损伤是否具有保护作用。方法 28只7~9周的清洁级ICR雄性小鼠,按完全随机分组法随机分为4组(每组7只):对照组(CON组)、机械通气组(VEN组) 、红景天苷组(SDS组)和红景天苷+机械通气组(SDS+VEN组)。VEN、SDS+VEN组机械通气4小时后或CON组和SDS组麻醉插管4小时后检测支气管肺泡灌洗液中(bronchoalveolar lavage fluid,BALF)蛋白浓度和细胞数量以及丙二醛(malondialdehyde,MDA)和超氧化物歧化酶(superoxide dismutase, SOD)的含量变化,测定肺组织湿/干重比(wet-to-dry weight ratio,W/D),观察肺组织病理改变,并用酶联免疫吸附试验(enzyme-linked immunosorbent assay,ELISA)检测肺组织中白细胞介素-1β(interleukin,IL1-β)、肿瘤坏死因子-α(tumor necrosis factor-a,TNF-α)的含量。结果 BALF中蛋白浓度和细胞数量:VEN组分别为(0.244 0.057)g/L和(2.02 0.33)个/ml,与CON组比较明显升高(P<0.01);SDS +VEN组分别为(0.170 0.023)g/L和(1.56 0.28)个/ml,与VEN组比较显著下降(P<0.05)。VEN组肺组织W/D(5.11 0.12)与CON组(4.69 0.08)比较,差异有统计学意义(P<0.01),SDS +VEN组肺组织W/D(4.77 0.05)与VEN组比较,差异有统计学意义(P<0.05)。SDS预处理能显著减轻通气诱导的肺组织病理损伤。机械通气4 h可诱导肺组织中IL-1β和TNF-α蛋白表达显著升高,SDS预处理可减轻机械通气诱导的IL-1β和TNF-α表达:SDS+VEN组(14.4 2.3)mg/g和(15.1 1.8)ng/g与VEN组(19.5 5.9)mg/g和(18.2 3.9)ng/g比较,差异有统计学意义(P<0.05)。与CON组相比,VEN组灌洗液中MDA含量明显增加,SOD活性降低,而SDS预处理则明显降低MDA含量,增加SOD活性: SDS+VEN组(1.89 0.32)mg/g和(41 4)ng/g与VEN组(2.57 0.60)mg/g和(32 5)ng/g比较,差异有统计学意义(P<0.05)。结论 机械通气前给予红景天苷可有效减少小鼠肺组织炎症细胞聚集,减轻肺水肿,机制可能与其抑制氧化应激水平相关。

Abstract:

【Abstract】Objective To investigate the effects and mechanism of salidroside on ventilator induced acute lung injury in mice. Methods tweenty-eight male ICR mice were randomly divided into 4 groups: group control (CON group,n=7),Mechanical ventilation group(VEN group, n=7), Salidroside group(SDS group,n=7),Salidroside +Mechanical ventilation group (SDS+VEN group,n=7). After 4 hours ventilation,collecting bronchoalveolar lavage fluid(BALF) and detecting the contents of protein,inflammatory cell counts,malondialdehyde(MDA),superoxide dismutase(SOD) in lung lavage fluid,the wet-to-dry weight ratio (W/D)and morphopathogical changes of lung tissue were examined.Lung tissues were homogenated to detect the Interleukin(IL)-1β and tumor necrosis factor-α(TNF-α) level by enzyme-linked immunosorbent assay (ELISA).Results The concentration of protein and cell counts in VEN group(0.244 0.057g/L and 2.02 0.33/ml) were much higher than CON group(P<0.01),and SDS+VEN group(0.170 0.023g/L) and (1.56 0.28/ml) were significantly reduced by comparing with VEN group(P<0.05). Wet/dry weight ratio were much higher in VEN group(5.11 0.12) than CON(4.69 0.08,P<0.01) and SDS+VEN group(4.77_+0.05,P<0.05). Pre-treated with SDS could significantly attenuate ventilator induced lung tissue injury. Four hours mechanical ventilation markedly evelated the IL-1β and TNF-α level in lung tissue and SDS could decrease the expression of IL-1β and TNF-α,there were significant statistical difference between SDS+VEN group[(14.4 2.3)mg/g and(15.1 1.8)ng/g] and VEN group[(19.5 5.9)mg/g and(18.2 3.9)ng/g ] of IL-1β(P<0.05) and TNF-α(P<0.05). After 4 hours ventilation,the MDA content in lung lavage fluid was increased significantly,SOD activity w as decreased(P<0.05) ,compared with the ventilation group(2.57 0.60mg/g and 32 5ng/g),the MDA was decreased and SOD activity was increased in salidreside group(1.89 0.32mg/g and 41 4ng/g). Conclusion Our results indicate that pre-treated with salidroside could significantly attenuate mechanical ventilation induced lung edema and the perfusion of inflammatory cells in mice, which might partly through inhibiting the oxidative stress level.
