盐酸右美托咪定对睡眠剥夺大鼠记忆和痛阈的影响

Effects of dexmedetomidine on the memory and pain threshold in rats with sleep deprivation

摘要:

目的 观察腹腔内注射盐酸右美托咪定(dexmedetomidine, Dex)对睡眠剥夺大鼠记忆和痛阈的影响。 方法 采用随机数字表法将90只健康成年SD大鼠分为3组(每组30只):对照组(C组)、睡眠剥夺组(SD组)、睡眠剥夺+Dex组(SD+D组)。SD组和SD+D组大鼠被置于盛满水的玻璃容器中央平台,每天10 h,持续8 d。C组大鼠被置于无水玻璃容器平台上,其余条件与SD组和SD+D组完全相同。SD+D组腹腔内注射50 μg/kg Dex,1次/d,C组和SD组腹腔内注射等容量生理盐水。每天Morris水迷宫定位航行实验测定大鼠逃避潜伏期,之后测定机械刺激缩足反射阈值(paw mechanical withdrawal threshold, PMWT)和热刺激缩足反射潜伏期(paw withdraw thermal latency, PWTL),持续8 d。 结果 与C组比较:SD组大鼠第3~8天逃避潜伏期明显延长,差异有统计学意义(P<0.05);SD+D组逃避潜伏期差异无统计学意义(P>0.05)。与SD组比较:SD+D组大鼠第3~8天逃避潜伏期明显缩短,差异有统计学意义(P<0.05)。与C组比较:SD组大鼠第2~8天PMWT明显降低,PWTL明显缩短,差异有统计学意义(P<0.05);SD+D组PMWT、PWTL差异均无统计学意义(P>0.05)。与 SD 组比较:C组和SD+D组大鼠第2~8天PMWT明显升高,PWTL明显延长,差异有统计学意义(P<0.05)。 结论 腹腔内注射Dex可以改善睡眠剥夺大鼠的记忆功能,减轻睡眠剥夺大鼠的疼痛。

Abstract:

Objective To investigate the effects of intraperitoneal injection of dexmedetomidine (Dex) on the memory and pain threshold in rats with sleep deprivation. Methods Ninety adult SD rats were randomly divided into three groups (n=30): a control group (group C), a group of sleep deprivation (group SD) and a group of sleep deprivation with Dex (group SD+D). The rats in groups SD and SD+D were placed on a glass platform within a tank filled with water, 10 h per day for 8 consecutive days. Meanwhile, those in group C were placed on a glass platform within a tank without water. Rats in group SD+D was intraperitoneally injected with 50 μg/kg Dex once a day, while those in groups C and SD were given the same amount of normal saline. Morris water maze positioning navigation behavior test was performed to measure escape latency every day. Then, paw mechanical withdrawal threshold (PMWT) and paw withdraw thermal latency (PWTL) were measured. Results Compared with group C, group SD showed obviously prolonged escape latency from day 3 to day 8(P<0.05). There is no difference in escape latency between groups SD+D and C (P>0.05). Compared with group SD, group SD+D presented obviously shortened escape latency from day 3 to day 8 (P<0.05). Compared with group C, group SD showed obviously reduced PMWT and PWTL from day 2 to day 8 (P<0.05). There is no difference in PMWT and PWTL between groups C and SD+D (P>0.05). Compared with group SD, groups C and SD+D presented obviously increased PMWT and PWTL from day 2 to day 8 (P<0.05). Conclusions Intraperitoneal injection of Dex can improve the memory of rats with sleep deprivation, and reduce the pain in those rats.
