仙茅苷对七氟醚麻醉老年大鼠术后认知功能的影响及其抗氧化和抗凋亡作用

Effects of curculigoside on cognitive function and its antioxidant and anti‑apoptotic effects in senile rats after sevoflurane anesthesia

摘要:

目的 探究仙茅苷对七氟醚麻醉老年大鼠术后认知功能的影响及其抗氧化和抗凋亡作用。 方法 30只大鼠按照随机数字表法分为两组(每组15只):围手术期神经认知障碍(perioperative neurocognitive disorders, PND)组和PND+仙茅苷组。两组大鼠根据参考文献建立PND模型,吸入七氟醚麻醉后进行腹腔探查。PND+仙茅苷组大鼠术后以仙茅苷(50 mg/kg)灌胃,每日1次,连续7 d;PND组大鼠灌胃等量生理盐水。记录大鼠水迷宫实验目标象限停留时间和穿越平台次数,H‑E染色检测海马组织损伤情况,ELISA法检测超氧化物歧化酶(superoxide dismutase, SOD)和丙二醛(malondialdehyde, MDA)水平,TUNEL染色检测海马神经元凋亡指数,Western blot法检测蛋白激酶B(protein kinase B, Akt)磷酸化水平。 结果 与PND组比较,PND+仙茅苷组大鼠目标象限停留时间、穿越平台次数明显增加(P<0.05),磷酸化Akt(phosphorylated‑Akt, p‑Akt)、Akt升高(P<0.05),SOD水平、Akt磷酸化水平明显升高(P<0.05),MDA和凋亡指数明显降低(P<0.05),海马组织损伤减轻。 结论 仙茅苷具有抑制氧化应激和海马神经元凋亡保护作用,改善老年PND大鼠的认知功能。

Abstract:

Objective To investigate the effects of curculigoside on postoperative cognitive function and its antioxidant and anti‑apoptotic effects in elderly rats. Methods Thirty rats were divided into two groups (15 in each group) according to the random number table method: perioperative neurocognitive disorders (PND) group and PND+curculigoside group. PND model was established according to the references, sevoflurane was inhaled into rats in PND group and PND+curculigoside group for abdominal exploration. Rats in PND+curculigoside group were given curculigoside (50 mg/kg) intragastric administration after operation, once a day for consecutive 7 days; PND group was given the same amount of normal saline. The target quadrant residence time and platform crossing times of the water maze were recorded, hematoxylin‑eosin (H‑E) staining was used to detect hippocampal injury, and enzyme‑linked immunosorbent assay (ELISA) was used to detect superoxide dismutase (SOD) and malondialdehyde (MDA) levels, the apoptosis index of hippocampal neurons was detected by terminal deoxynucleotidyl transferase‑mediated dUTP‑biotin nick end labeling (TUNEL) staining and the phosphorylation level of protein kinase B (Akt) was detected by Western blot. Results Compared with PND group, the target quadrant residence time, the number of platform crossing were significantly increased (P<0.05), hippocampal tissue damage was reduced, phosphorylated‑Akt (p‑Akt) and Akt were increased (P<0.05), SOD level and Akt phosphorylation level were significantly increased (P<0.05), MDA and apoptosis index were significantly decreased (P<0.05) in PND+curculigoside group. Conclusions Curculigoside can inhibit oxidative stress and protect hippocampal neurons from apoptosis, and improve the cognitive function of elderly PND rats.
