HMGB1促进RSV感染的THP⁃1单核细胞中NLRP3炎症小体的活化

摘要:

目的 探讨高迁移率族蛋白B1(high‑mobility group box 1, HMGB1)对呼吸道合胞病毒(respiratory syncytial virus, RSV)感染的THP‑1单核细胞中核苷酸结合域样受体蛋白3(nucleotide‑binding oligomerization domain‑like receptor protein 3, NLRP3)炎症小体活化的影响。 方法 常规培养THP⁃1细胞用于RSV病毒的感染实验,将培养的THP‑1单核细胞分为4组:未感染RSV组、RSV感染组、RSV感染+人重组HMGB1(rHMGB1)组、RSV感染+甘草素组,RSV感染6 h后继续培养48 h,提取4组细胞总蛋白,Western blot法检测4组THP‑1单核细胞的NLRP3、凋亡相关斑点样蛋白(apoptosis‑associated speck‑like protein containing CARD, ASC)、胱天蛋白酶‑1(cysteine aspartate specific protease‑1, caspase‑1)、IL‑1β水平。 结果 RSV感染组NLRP3、ASC、caspase‑1、IL‑1β水平高于未感染RSV组(P<0.05);RSV感染+rHMGB1组THP‑1单核细胞NLRP3、ASC、caspase‑1、IL‑1β水平高于RSV感染组(P<0.05);RSV感染+甘草素组NLRP3、ASC、caspase‑1、IL‑1β水平低于RSV感染+rHMGB1组(P<0.05)。 结论 HMGB1可通过促进RSV感染的THP1单核细胞中NLRP3炎症小体的活化进而促进IL‑1β的表达而致炎。

Abstract:

Objective To explore the effect of high‑mobility group box 1 (HMGB1) on the activation of nucleotide‑binding oligomerization domain‑like receptor protein 3 (NLRP3) inflammasome in respiratory syncytial virus (RSV) infected THP‑1 monocytes. Methods THP‑1 cell line was routinely cultured for infection with RSV virus. The cultured THP‑1 monocytes were divided into four groups: a non‑RSV infection group, a RSV infection group, a RSV infection plus rHMGB1 group, and a RSV infection plus glycyrrhizin group. After RSV infection for 6 h, the cells were cultured for another 48 h. Then, their total proteins were extracted. The levels of NLRP3, apoptosis‑associated speck‑like protein containing CARD (ASC), cysteine aspartate specific protease (caspase‑1), and IL‑1β were detected by Western blot. Results The RSV infection group showed significantly higher levels of NLRP3, ASC, caspase‑1, and IL‑1β than the non‑RSV infection group (P<0.05). The levels of NLRP3, ASC, caspase‑1 and IL‑1β in the RSV infection plus rHMGB1 group were significantly higher than those in the RSV infection group (P<0.05). The levels of NLRP3, ASC, caspase‑1 and IL‑1β in the RSV infection plus glycyrrhizin group were remarkably reduced, compared with those in the RSV infection plus rHMGB1 group (P<0.05). Conclusion HMGB1 can cause inflammation through promoting the activation of NLRP3 inflammasome and then inducing the expression of IL‑1β in RSV infected THP‑1 monocytes.
